Act 7
.. I recommend following this skill tree path which was explicitly designed to make the gaming experience as pleasant as possible for new players. That being said, it focuses more on survivability and comfort than damage since these are way more important at the beginning, but the guide will elaborate on options to adjust the skill tree in order to be more damage centric for the endgame later.
Pantheons are strong permanent passive effects your character gets by defeating specific bosses. Here, you can choose the minor Pantheon ‘Soul of Ralakesh‘ after killing ‚Ralakesh, Master of a Million Faces‚ in The Ashen Fields in Act 7 by talking to Sin who provides you the following buff: “25% reduced Physical Damage over Time Damage taken while moving + Moving while Bleeding doesn’t cause you to take extra Damage”. This Pantheon is not your minor endgame Pantheon, but the best choice for now.
You will do The Lord’s Labyrinth soon, so there’s no benefit in skipping this now. Trials of Ascendancy are mazes within specific areas of the campaign – in this case, The Crypt.

Do this now since you will do The Lord’s Labyrinth next. You can find all trial locations in Act 1-7 here.
You can find the Aspirants‘ Plaza in The Sarn Encampment in Act 3. It is accessible through the Statue of the Goddess (at the top of the stairs).
Once you’ve completed The Normal Labyrinth, you can choose your Ascendancy class which gives you access to your Ascendancy skill tree. You have to choose the Champion Ascendancy class. Your first two Ascendancy skill points are going to be invested in Unstoppable Hero.
You can find the respective option at the end of the room on the top right side (don’t go through the portal before you’ve chosen your Ascendancy class – otherwise you have to redo the whole Labyrinth).
Note: The layout of The Normal Labyrinth changes on a daily basis. You can find the optimal pathing for the current day here.
Once you’ve completed The Cruel Labyrinth you can invest 2 additional Ascendancy skill points. Here, you want to go for Fortitude next.
